I first add the isopro backpacking stove fuel, then add the propane to the system. I charge while the system is running, charge as liquid after agitating the can of fuel to make sure it's well-mixed. An old R12 can top tapper will usually fit on the isopro cans if you want to go that route. An R134A can tapping valve will not.
The ideal mixture is somewhere around 75% propane and 25% isobutane by mass. You should be able to do the math to figure out how much this is. I would put in about 12oz of gas. You can weigh the gas cans before and afterwards. For MSR isopro, the math is 3.75oz of isopro, then 8.25oz of straight propane. I am never so quantitative about it. I usually put in about a half can of isopro, then top it off with bbq or bernzomatic propane to get the low side pressure up into the 35-50psi range on my manifold gauges, depending on the outdoor ambient temp. There should be bubbles in the sight glass. Or maybe even half gas half liquid in the sight glass. Too little is better than too much in this game.
For leak detection, like I said, I just use soap.
