No gasket is needed, you do not need to pull the intake manifold. It is really fidgety in the back, with the ECM, a bunch of wiring, and some vacuum pipes back there. The vacuum pipes are prone to break - either from you being there, or age, or both. Be prepared for them to be broken. They are not cheap, but I would consider just replacing them if they haven't already been. I believe the GM part numbers are 12588555 and 55556698 - one is part of the PCV system, the other part of the brake booster. Similarly, you're going to want to replace the spark plugs unless they are brand new. You don't want to go in there twice.
Misfires should be detected readily by the ECM, as should knock. It's possible you have weak coils but IMHO anything serious enough in-cylinder to produce an audible noise should set a code. I'd probably look elsewhere. Still, if the car has more than 100k on it, this work will come up sooner rather than later, no harm in tackling it in advance. I did the same.
