Do you notice the transmission 'hunting' - that is, shifting gears seemingly unnecessarily? If it's doing that, that's (1) annoying, and (2) will increase wear, somewhat.
But so much depends on the transmission and the software driving it. When OD autoboxes started showing up in the '80 in bulk, there were a bunch of cars, notably domestic, which would hunt in the 40 mph range as they kicked in and out of OD. In the intervening 20 years, designers have both the luxury of computer-controlled automatics, and a few more years (decades) of experience. So I think your co-worker is warning you against something that doesn't happen anymore, but was a concern In The Old Days. Remember when turbo equalled grendaded engine?
If you aren't noticing any transmission hunting, stick it into D and leave it there. And don't go looking for it, either. A major disadvantage of putting it in 3rd is forgetting it's there - and I'll bet you hit 45 mph a bunch of times in the city. And if you forget and get on the highway, with today's well insulated cars, it may be a while before you notice that the tach is sitting at 4500 rpm.
