Yes, the cap for the reservoir. The reason I suggested this is that based on your symptoms it seems to be pressure related rather than dependent on flow or simply leaking all the time. It is easy and cheap enough to try that you don't really risk anything by switching the cap. With the cover over the reservoir it can be hard to see, especially since the cap vents out the bottom. You might pull the cover off so you have a better view of what's going on.

Not sure I understand your other question - are you suggesting it might be something other than coolant (i.e., transmission fluid), or are you asking if a coolant leak might result in transmission damage? If the latter, the answer is no - an automatic relies on the coolant somewhat, while a manual doesn't use it at all, but the engine would overheat much sooner in either case.
