Track down the coolant leak first. The most likely sites are the weep hole on the water pump or a leaking hose (the small elbow on the front of the engine is a good candidate). Could also be a rusted freeze plug or other locations.
Lots of possibilities for the running problem, including a bad head gasket. Eliminate the simple items first, like bad spark plug gap, rotor, distributor cap, plug wire, fuel injector, etc. If you've got a timing light, hook it to the cylinder you think is bad and then start the car from the condition that leads to the cylinder not firing. If the light flashes steadily despite the fact that the cylinder is missing, then the ignition system is OK, and the problem is elsewhere (fuel or plug gap, etc.). If the light flashes eratically or not at all until the cylinder starts to fire properly, then the problem is w/ the ignition.
