First - what exactly is your problem with the car? If you are losing fuel pressure due to the check valve the test is easy. Get a fuel pressure guage (it must be one designed to work with FI systems - the one you describe will not read high enough) - 0-5bar is a good range. Connect it via a T adaptor between the fuel rail and the FPR ( you may have to do some cutting and joining but make sure it is a good job as fuel is at a high pressure) and run the engine. With the vacuum hose disconnected you will see if the FPR is within limits (the value is stamped on the unit)
When you switch off the engine the pressure should drop a bit (see the manual) but still retain some pressure. If it falls right off you have a dodgy check valve. As far as the various combinations of engine off pump on are concerned I'm pretty sure it won't make any difference to the tests. If you feel safer doing the tests without the engine running you can jump the relay with some wire (sorry I have forgotten the pin numbers though. (sorry if I'm stating the obvious on a few points)
