I'm assuming that the engine/trans is still out of the car.
With the transmission out of the car, you need to move the shifter as far to the right and as as far back as it takes to engage the lockout lever. Move it slowly and you should be able to feel where it's contacting the lockout lever (and hear it, if it's really quiet). It's pretty subtle, but it's doable.
Installing the lock cylinder is pretty straighforward, so you can't do much wrong. If it turned before you put it in, it should turn now. Did you do anything with the gears that connect the cylinder to the switch?
