You should not be touching the engine control unit (ECU) for this. The central lock control unit is a different and separate device.
That said, perform an electrical check on the switch on the driver's door lock cylinder. Mine's getting old and sometimes does not register.
If that works, try pulling fuse 'A' under the seat (The one that controls central locking) for ten seconds or so. Sometimes the control unit on my car gets confused after repeated lock/unlock cycles and will stop working for ten seconds. Once it quit working altogether (the switch was fouled and sending the wrong signals) and pulling the fuse let it sort things out.
