It's probably a big deal. I'll bet that if you listen, you'll hear a whine in 3/4/5 above ~30 mph when under load. If so, that's the pinion bearing. You could have some other bits in there failing too, none of which is good. These types of problems do not cure themselves and if you let it go, you'll just cause more expense when it comes to fixing the tranny.
Hope you've got a big piggy bank because you're in for a tranny rebuild/replacement. Don't drive it.
