Usually you would undo the fitting at the end with the hard brake line. THat fitting should turn on the line. The other end screws straight into the caliper.
But before you do that, remove the bleeder screw completely, and clean it out. Often they get filled with crud (technical automotive term, that) and the fluid can't get past it. See that you have fluid coming through the socket for the bleeder screw when someone presses on the pedal, or you operate your pressure bleeder.
