Yes, you can pull the check valve straight up. But with a LOT of caution-

First, depending on the year, there is a retention clip on top of the valve. On older cars, there is a cap that is held down with a screw. On newer cars, there is a little yellow plastic clip that has to be pulled back. I just did my fuel pump, and I took some carpet thread and tied the clip back just enough for the valve to clear.

Start the car and pull the fuel pump fuse until the car dies. This will depressurize the fuel system some. The valve is amazingly fragile. Give it a very slight twisting motion while gently pulling up. Pull up ONLY on the vertical portion; if you pull up on the hose side, it will snap. The valve has two O-rings that hold it in place. When I took the valve out, I twisted (not a lot, maybe 15 degrees side to side) and pulled up gently, and worked at it for at least five to ten minutes.

I have a silly question - is the valve barb (plastic piece) itself leaking, or is it where it meets the tube? Could you just slip a clamp over the tube, and avoid pulling the valve out at all?

If you do end up replacing the valve, from various posts I've seen that it is a pain to get the new valve into the hose. Some folks have used a heat gun to keep the hose soft, and used a brake flaring tool to expand the hose enough to replace the valve. Apparently it can't easily be done by hand. Look back through the archives on "fuel pump", and you'll see a lot of talk about those fuel valves.
