As Ari notes, plugs are a likely culprit, or the DI is unhappy.
However, my '96 has started on 2 or 3 cylinders twice in the last 30K miles. The first time it went away after about 15 seconds, the second time it took a couple minutes. In each case the plugs were recent (well under 10K miles). I never changed a thing, or even checked them as the car ran fine once the missing stopped. And it has run fine since.
In my experience with fuel injected cars, they will sometimes randomly misfire on start-up, then all will be well. Is it misc gunk in the fuel system, a little plug fouling or something else? I dunno, but in a '78 Scirocco, an '86 GTI, and '84 C900, my wife's (now departed Exploder) and my '96 9000 this has happened to everyone of them. Probably has on my daughter's '95 9000, too. On some cars it can be more common when the engine is hot.
If it recurs you have an issue, but a random start-up misfire seems to be the norm, in my experience, as long as it goes away in a minute.
