The alignment marks on the cam can be used as a wear indicator. One indication of wear is the chain getting longer. If the marks on the cams are 'outside' the marks on the head - then the chain is effectively being measured between the fixed points of the cam. Chain wear is thousandths of an inch on each pin and link hole, added up over the length of the chain. I don't think you could place the guides accurately enough so that the measurement would be meaningful. Also, you're trying to measure the chain, not guide wear, but the two measurements would be intermingled.
I'll stick with the alignment marks as a measure of wear, along with good old visual inspection. I've never been a big believer in the chain tensioner extension method. I think it's too open to mistakes when removing the tensioner. It is good as a quick and dirty test that requires very little labor, and is therefore excellent for a shop, especially since the downside (gee, you need a new chain) isn't that bad for the shop.
