On the top or in the trunk?

I'm going on a long road trip and am debating whether I should cram all my gear in the trunk or in a roof box. I have a Thule Evolution 16 box (fairly aerodynamic)... and it would make life much easier to put stuff up there so I can leave the backseat and trunk relatively empty. The trade-off of course would be the loss in gas mileage having a box on top.
Does anyone have experience with how much a roof box actually affects mileage? It has occurred to me that having the weight up there and therefore centered over the car might be better for handling anyway.
Thoughts? Suggestions?
