If you see a hole in a muffler, chances are good that the metal around the hole is just about ready to fail. So most 'plugs' don't work, because the surrounding metal fails because plugging the hole increases the pressure in the area. It would be different if the hole were due to a puncture, but they're usually due to rot. And due to surface corrosion, any adhesive is trying to stick to material that is about to flake off. You could go in and clean up the surface - grind it down to bare metal - but you'll find the hole is a LOT bigger all of a sudden.

I'd go with what the others have said - muffler wrap or muffler bandage. you wrap the area with a heat-resistant material impregnated with a cement-like goop. That provides the sealing. And then it gets wrapped with wire or metal tape - that provides the mechanical support. If the hole is in the end of the muffler, a wrap won't work very well.

But any 'fix' is just temporary. I've been able to limp a muffler-wrapped resonator along for a couple of months, but it wasn't pretty (or particularly quiet). Mufflers don't just develop one hole - viewed from the inside it's a mass of rust, and the hole you see on the outside is just the beginning. So I'd go with muffler wrap (or a beer can and bailing wire), and go online to order a new muffler.
