I've found that the paid, active version of Malwarebytes working in conjunction with antivirus is very effective.
FWIW, MS Security Essentials in my experience is not particularly good at blocking malware, its main benefit is that it is very lightweight and won't slow the machine down so it's nice for older systems whose owners are not high-risk users.
In my experience all comprehensive "internet security suites" are extremely invasive and cause stability and performance problems. I always advise clients to go with a solid, basic antivirus package. (Most of my people are using Kaspersky AV.) Many times I've been asked to work on a Windows system with severe performance problems and it comes alive when [Norton/McAfee/whatever] Internet Security Suite is uninstalled.
For those wanting free antivirus lately I've been installing Panda Cloud Antivirus.
A lot of course depends on usage patterns and random chance. (People sometimes ask what I use for antivirus and the answer is "none" -- I run Linux and use Noscript and other browser plugins to maintain control over what goes on.)
The important thing to remember is that security is not a program that you install on your PC, security is a process. A multi-layered defense strategy is required, the first layer being your own brain!
