It IS normal not to be able to 'back-drive' the window regulator and motor. In the little gearbox inside the motor assembly there's a worm gear on the motor shaft that drives a large nylon gear that's on the same shaft as the little pinion on the outside of the gearbox (the one that drives the regulator quadrant). Worm gear arrangements like this have so much mechanical advantage one direction that it's not possible to drive them the other way. It'd be like trying to spin a bolt by pushing or pulling on the nut!
With the motor assembly removed from the regulator, you can move the window up and down by hand. There's no other 'locking' feature, but there is a counter balance spring in the regulator.
As RS's post points out, you _can_ get the thing apart with the window up. I had to do it a couple weeks ago, and probably did it just as RS describes - completely disconnect the regulator from the door (5 bolts?), then start wrestling. Eventually I was able to get access to the two screws that hold the glass on.
