The red/white check valve goes so that air (vacuum) can be drawn into intake manifold.
Usually you don't need to calibrate after those two parts, but if you continue to have problems, a calibration is one of the first things to do to make sure that that is not the cause of the problem.
You can remove most of the slack in throttle wire with that adjuster. With a perfectly running TCS system, that wire is limp any way as soon as the engine starts and the vacuum from through bleeder valve pulls the throttle diaphragm open.
