Compressed air does work, but you have to be careful. The drain hoses are held on with clamps, and I've seen the hoses come off all by themselves. Too much air, and you could blow the hose off the drain pan nipple. Not too hard to fix - remove the rim trim around the headliner and pull the headliner down a little to re-attach.
I'd try air first just keep the pressure low. What's low? Damn fine question - let us know.
As to the rubber nipples - they are the most common place for the clog. Why have them? I have to figure that if you have the sunroof open and there is no nipple, the venturi effect could cause the hose at the bottom to suck up some road sand. I remove the nipples to clean, and then replace them, when I remember. Of course, they do tend to fall off over time.
I like using a flexible, soft wire. I've used 16 gauge insulated STRANDED electrical wire, just making sure the business end is blunt - you don't want to poke a hole through the hose.
