I would test drive the car again and check. With the misfire, the engine feels like it's studdering, almost as if it's being held back. If you notice that at the upper end of the yellow, then I would feel comfortable that the problem is nothing more than the spark plugs. It's very common.
If it were a power transmission problem with the tranny, you'd notice it worse in 1st and 2nd gears, as that's when you get the most torque through the box. Frankly, tranny problems show up as late or harsh shifts. Unless the tranny is slipping (revs increasing more than road speed), I'd say it's highly unlikely it's a tranny problem.
Heck, see if you can sell the problem to the seller it's a tranny problem and use that to bargain the price down. THe spark plug gap increases with age, and old, poorly-gapped spark plugs show up as misfiring at high boost in turbos. That's what you're seeing.
