Of course, disconnect the negative lead of the battery. You'll be doing most of the job by feel, and don't want to slip and cause a major arc.
I assume you're going in through the RF wheel well. I ask because some folks try it from above. I use the wheel well access.
Sometimes the screws holding in the regulator come out nicely, sometimes they don't. If they start to become a real problem, bite bullet and pull the alternator. If you really mess up the screws, it can be hard to get them out even with the alternator on the bench.
Make sure you know EXACTLY where the thin wire (yellow?) coming to the back of the alternator goes. Put that back in the wrong place, and things won't work.
