It probably wasn't a good idea to mess with the mixture screw because it ran fine a few weeks ago- and the odds are the screw didn't suddenly move on it's own. If you don't remember how many turns you adjusted it, just turn it all the way in and then out 1 1/2 turns. This will be a good starting point. Is the manual choke cable stuck in the closed or semi closed position? If not, read on-
Step one to get a stoker running right is a complete tune up. Stokers need complete tune ups every 3,000 to 6,000 miles, depending on how good your luck is. I tune up every 3,000 miles because I hate getting stranded.
You should replace the plugs with NGK A-8's, A-7's are too hot and could hole a piston. Replace the points, distributor cap, condensor and rotor. After replacing these parts, make sure your timing is dead on. Static time it (engine off), the points should spark (snap) at 7 degrees before tdc. Change the gas filter. It the car still runs badly after the tune up it's time for a compression check. If you have low compression in one cylinder- bummer (you know what that means).
From my experiance with strokers, nine out of ten times the problem is in the ignition system. It's almost never a fuel feeding problem (fuel pump) or a carb problem. But who knows? Try the tune up first and see what happens.
