Typically one can re-use a self locking nut (ie one with a nylon insert) if the running torque is at or above 5 ft-lbs. The running torque being the torque value while turning the nut prior to clamping load being engaged. The nylon insert will generally produce these values 4 or 5 times before expiring. The nylon insert prevents the nut from backing off once the joint pre load is lost. A properly torqued fastener (ie one that has stretched the thread materials to about 80 percent of their yield strength should never loosen anyway. Those fasteners used in applications subject to significant vibration may get torqued close to 100% of yield.
