You can detect a hole in the diaphragm by using your mouth to develop suction via the hose barb. A minor leak would be of no consequence, but can be detected by a leakdowm test by sealing the hose barb with your lip.
You can press the valve plug up as high as it will go, and then close the hose barb with your finger and then release the valve plug. A vacuum will be created and will be sustained while the hose barb is closed by your finger.
Either of the above will qualify the diaprhagm. You have already determined that the valve seals while seated.
A bosch valve in good order can become very musical with higher boost pressures. I and others have replaced with piston type valves with the primary objective being to stop the tooting.
The bosch hose barb needs to point to the battery, but it might be quieter if flip-flopped. I found that washing mine to remove the oil worked very well at reducing noise... for a while.
