Couple of options:
Northern route: if you start directly from Whistler, you'd be continuing north via Hwy #99 to Cache Creek. From C.C., go on Trans-Canada Hwy (aka TCH or Hwy #1) and head east towards Kamloops. Whistler to Kamloops leg is approx. 3 to 4 hrs. From Kamloops to Banff via Hwy #1 is another 4-5 hrs depending on traffic (esp. RVs). Total time up to 9 hrs.
Southern route: if you were to come over to Vancouver instead, figure about 2 to 2.5 hrs to get into town via the ferry. Then another 1.5 hrs from Vancouver to Hope via Hwy #1. From Hope to Kamloops via Hwy #5 (Coquihalla Hwy, $10 toll) is another 2 hrs. Then from Kamloops to Banff via Hwy #1 to Banff is again about 4-5 hrs depending on traffic. Total time up to 11 hrs.
These are based on my own driving experiences. Either way, two full days would ensure a nice leisurely pace if you camp or B&B along the way. Mind you there are numerous forest fires in BC right now so expect road closures, delays, and hazy conditions. Check roads status at:

A good halfway point between Whistler and Banff along the northern route would be near the town of Clearwater and Wells Gray Provincial Park (about 1.5 hrs north of Kamloops along Yellowhead Hwy #5). It's a bit of diversion but the scenery is spectacular inside the park and there's lots of ranch houses to stay at. We just came back from there in July and we stayed at the Wells Gray Guest Ranch. Tons of outdoor activities, fun for all.
