I've flown 40-50K a year for the last 5 years and will hit almost 60K this year. Here's what works for me.
Noise canceling headphones.
These are truly wonderful devices. It's amazing how less tired you are after flight (especially a 9 or 10 hour one) if you can limit the cabin and engine noise. You are NOT limited to Bose, which are overrated, overpriced and much too bulky. My wife got me a set of Plane Quiet NC-6 phones for Christmas last year and I've flown with them over 50K miles so far this year and they are great. (See link below) The newer versions, NC-7, are supposedly even better. They are about a third the price of the Bose ego-enhancing units. Sennheisers are also very good, and much less than Bose.
Music
For tunes I prefer an iPod. I have a 30GB model and the battery lasts for about 12 hours, which will get you to most places you're likely to go. All are good. Get whichever one floats your boat. Plus, in a pinch you can actually carry some work info in a file on it and pull it over to your laptop if needed.
Earplugs
When you don't want music, need to sleep, etc., a set of silicone earplugs works wonders. Also great if you end up on some prop-driven puddle jumper, because they tend to push the limits of headphones.
Airline club
If you will be travelling a lot consider joining an airline club. I did it this year after years of resisting. Worth every cent. Pick the one based on the airline you'll be using the most and where you have to go. When your flight is delayed (normal) the terminal is a zoo and you want someplace comfortable to sit and call home/office or just work, a club is a real refuge. Comfy chairs, bar, TVs.
Worth considering
Neck pillow. Not everyone likes these, but they are nice on long flights, especially if you are stuck in coach and want to sleep. I got a good one at Brookstone for about $25. I only take it on cross-country or transoceanic trips, but great to have.
Finally, stay in decent hotels.
