The entire metal case of the alternator should be ground; the only positive points on the alternator are the main power bolt, and the smaller bolt where a single wire is hooked up that leads back to the battery light on the dash. So literally find a good metal point on the engine (throttle body should work well) to use, and do the following:
1. set your multimeter to read Volts on an appropriate scale (I use 20v on my digital multimeter)
2. put the negative lead on the engine (sometimes you might scratch a bit to be sure you have good electrical contact
3. put the positive lead on the alternator case (scratch for good electrical contact here as well.
4. Read the difference. If the alternator is properly grounded, there should be little or no voltage across the connection; if not, there may be a significant voltage difference.
