which car we are working on so I will give you the basic answer to your question. The wastegate lines aren't really the best or easiest line to use and actually doesn't give the best results. You want to get to the "log" on the manifold. Late cars have a port right next to the oil filler cap - you remove the rubber plug and put your line there. this now takes in to all account losses, all losses in the induction pipe, losses across the throttle body and you are seeing precisely the same pressures your ECU isw seeing - that is what you want to measure. The alternative is to tee into the fuel regulator line or the line to the Absolute Pressure Sensor (MAPS)
Disconnect the speed sensor if you have trionic control or the electrical connector to the Turbo Boost Solenoid and drive the car as normal you will reach the base boost. If you apply brakes this will affect the reading on DI cars because the advance is retarded to Reference. Max boost is with your car running with everything hooked up. then drive it through the gears and record the highest boost achievable. some times theis is best done with a co-driver reading the gage and recording. What I like to do is atke the car out on a good road and shift to 4 around 40 (70K) and floor the car and have the co-driver read and record depending on your car turbo you reach max boost at slightly different RPM> Record the point where base boost comes in from your previous test this RPM will be used later for reference in testing. continue to accelerate until 3500-4000rpm-4500 T25 turbos. This isn't very scientific but it does work and you have a thrill doing it! This will be pretty near the max boost for most HOT cars. at least so close any beyond this will be too fast to measure. If you are restricted by the road you may try 3rd but thing happen so fast it is difficult to record and dynamics come into play. If you have access to a dyno that would be perfect but expensive for what you are trying to acomplish. usually if one sets the base boost correctly the max will take care of its self but when you go to playing with base boost and clamping the wastegate tighter is when thing tend to go wrong, especially with trionic cars.
