How is the lock action with the knob on the inside? If the action is smooth both ways, then yes, it's probably the lock cylinder, or its connection to the rest of the lock mechanism. Even if it's the same, it sounds like time to open up the door and root around all the levers and bars and assorted "Rube Goldberg" bits that make up the locks inside. You've got a short lever on the lock cylinder that acts to move the same lever that the plastic knob does on the inside, which also is connected to the switch for the central locking, and someplace in all that, actually connects to the latch mechanism, so there are several potential points of problems inside the door. Only by getting in there will you find out what's what.
