First, confirm it is not the speaker, which I think you have done. If not take the wires from the working speaker and connect to the suspect unit. If the speaker is operative than you must go upstream.
I would try to pull the head unit and check output to the suspect speaker. If you read a signal than the wire inbetween the unit and the speaker may be shorted, if no signal you have a dead channel output in your head unit.
I have never pulled my radio and I suspect one of those block connectors, so you will need a wiring diagram to figure out which color wire represents the specific channel you are trying to test.
