Always disconnect the ground (negative) first. That insures that if the wrench inadvertently touches something, there will be no current flow (no sparks) because the negative terminal and the chassis are electrically connected (until you remove the cable from the battery)

You can then remove the positive terminal from the battery without fear of shorting between the terminal and the chassis (or the AC plumbing) and causing sparks and possible damage to things. With the negative disconnected, there is no way to complete a circuit from the positive terminal through any part of the car.

If you want to check for current drain on the various circuits, it is easier to leave the battery connected, and check at the fuse/relay box. Pull a fuse and touch the leads of the VOM (set to read amps at the highest scale) in the two legs of the socket from which the fuse is removed. If the circuit is drawing current, the meter will show it. If needed, switch to lower amperage setting to check for very small current flows. The clock/radio should draw a few milliamps.

Note: IIRC the ignition/starter circuit doesn't go through the fuse box, so you'll have to test for that at the battery.
