Using teflon tape on the treads won't do much to seal things. The seal is made by the flange on the end of the tubing seating against a mating part of the other half of the connection. The teflon tape could actually impede a good seal if it gets in the wrong place and may keep the nut from seating fully. I would recommend removing the teflon tape and starting with clean threads.
As for bleeding the clutch, I've had the best luck by injecting hydraulic (brake) fluid into the bleed nipple at the slave cylinder. This pushes the fluid (and bubbles) up toward the reservoir, in the direction that the bubbles naturally tend to move. Use a length of hose and a syringe, turkey baster, or just a funnel to feed the fluid into the system. Keep the fluid source above the level of the master cylinder to prevent air bubbles from siphoning back into the system.
If you keep getting air bubbles in the system, either the system has a leak, or air is getting back in as you tighten the bleed nipple. This can happen when you disconnect the hose leading to the bleed nipple from the vacuum or fluid source and then let the free end of the hose drop. As fluid flows out of the free end of the hose, air will siphon in at the bleed nipple. To avoid this, keep the free end of the hose (full of fluid) up above the level of the master cylinder and twist the hose to do the preliminary tightening of the bleed nipple. Pull the hose off and quickly finish tightening with a wrench.
