I think the most important question that hasn't been asked yet is how you're going to drive to a city that is only accessible by air or water ;). Obviously you'll be catching a ferry somewhere.
All the suggestions posted so far have been good but I dont think it's really going to be that rugged, depending on which port you leave for Juneau from and which route you take to get there. The climate on the coast is fairly mild. Your departure point (SD) is going to be vastly colder than your destination. I'm sure you already know how to take care of yourself in the cold.
I have driven the Al-Can hwy several times (Anchorage, AK -> Seattle), however never in the winter. The entire length of the Al-Can is plowed but I dont know about other highways in Canada. I never ran out of gas. Much of the hwy was being worked on at this time (as always) and I got a few flats. There was always a repair facility within ~ 100 miles. This is just my experience. The route you take may very well be more desolate than mine. Its obviously best to be well prepared. Better to have and not need, than to need and not have.
