The nut came loose because it was not tightened properly at some point and time. They never come loose otherwise.

The goo and sealer on top of those upper threads won't really do much.
It is the lower part of the thread going into the transmission casing that all suggestion are about when talking thread sealer. But for that you would have to extract the stud completly out of the transmission casing which is a whole 'nother thread post in itself.

Also, if that nut is not tight enough, it will be motor oil seeping out between the engine and transmission casing.
One problem if that has already happened is that the bond between the casing gasket and the silicon used at the time will now have a breach and it might be difficult to make it seal again.
So it is of high importance that you get the nut tight and also check the other bolt going in from under, closer to the alternator side so that that one is tight too.

Back in the old days with the B-engine, it used to be a paper gasket between engine and transmission and on the turbos it blew out creating an oil leak. That's when Saab started to use the sheet metal gasket instead with grooves for extra silicone at the ends.
