I would run the car with the valve cover off and very carefully verify that the noise is from the lifter. As suggested, make sure you don't have a exhaust manifold or intake manifold leak. If bad lifter is verified, then replace the bad lifter. Not really a bad job. If you tie the cam sprockets to the timing chain in position with tie strips, you will not mess up cam timing.
Your bigger issue is timing chain related. There is a good chance that the guides are worn, and a smaller chance that the sprockets are worn. While a roll-in chain will help and is a good fix if guides are OK, worn guides could reduce longevity. Also, if the sprocket teeth are worn at all, the chain will very quickly go bad. The lower sprocket on the crank is the PITA, as are the chain guides. All can be done with the engine in the car, but the task is daunting. Much easier with engine pulled.
